Xgeva (denosumab) is a fully human monoclonal antibody that targets and inhibits RANK ligand (RANKL), a key protein involved in the formation, function, and survival of osteoclasts, the cells responsible for bone resorption. By blocking RANKL, Xgeva helps reduce bone destruction and strengthens bone. It is prescribed for the prevention of skeletal-related events (such as fractures, spinal cord compression, or the need for radiation or surgery to bone) in patients with solid tumors that have metastasized to the bone. Xgeva is also indicated for the treatment of adults and skeletally mature adolescents with giant cell tumor of bone that cannot be surgically removed, and for patients with hypercalcemia of malignancy that does not respond to bisphosphonate therapy. It is given as a subcutaneous injection, usually once every 4 weeks.
